科研项目

1、本人于2016年至2019年间,作为项目负责人承担了国家自然科学基金面上项目(项目编号81573561),该项目题为“基于GLUT4和SGLT2双靶点的槐属药用植物抗糖尿药效物质基础及作用机制研究”,获批经费总额为66万元。2、在2014年至2017年期间,本人作为项目负责人主持了武汉市国家东湖高新区光谷创新人才计划项目(项目编号BX219),该项目致力于新药中药六类川七镇脑宁胶囊的开发研究,获得项目资助经费100万元。3、本人于2014年至2015年负责了中国科学院上海药物研究所新药研究重点实验室开放基金项目(项目编号SIMM1403KF-07),该课题为“基于GLUT4靶点的广豆根抗糖尿病活性物质和作用机制研究”,获得资助经费8万元。4、在2013年至2014年间,本人作为项目负责人承担了武汉市科技计划项目青年晨光计划项目(项目编号2013070104010028),该课题主要进行苦参抗糖尿病有效部位的临床前药效和作用机制研究,项目经费为10万元。5、本人于2012年至2014年主持了国家自然科学基金青年科学基金项目(项目编号81102798),该研究聚焦于基于GLUT4靶点的苦参抗糖尿病活性物质和作用机制,获批经费22万元。6、在2012年至2014年期间,本人还负责了人事部留学回国人员择优资助项目(项目编号BZY12006),该课题同样围绕基于GLUT4靶点的苦参抗糖尿病活性物质和作用机制展开研究,获得资助经费3万元。7、本人于2013年至2015年作为项目负责人承担了中央高校基本科研业务费专项资金资助项目(项目编号3212014CZW14051),该课题致力于藏药马先蒿质量标准的研究工作,项目经费为5.5万元。8、在2011年至2013年间,本人主持了中南民族大学人才引进基金项目(项目编号YZ211002),该研究主题为基于GLUT4靶点的苦参抗糖尿病活性物质和作用机制,获得资助经费90万元。9、在“十二五”期间,本人作为科研骨干参与了国家科技支撑计划项目(项目编号2012BAI27B06),该项目题为“民族医特色疗法疗效评价及平台技术研究”,执行期为2011年至2015年,本人分配到的研究经费为40万元。10、本人于2012年至2014年参与了青海省自然科学基金重点项目(项目编号2012-Z-904),该项目主要进行藏药预分离组分库建立与抗肝癌活性研究,本人排名第二,分配经费10万元。11、在2016年至2018年期间,本人再次参与了青海省自然科学基金重点项目(项目编号2016-ZJ-908),该课题致力于基于Caspase 3靶点和预分离组分库的藏药抗肝癌药效物质及其作用机制研究,本人排名第二,分配经费10万元。

学术研究

本研究致力于从天然资源库中分离、纯化并鉴定具有生物活性的天然产物,并在此基础上,以葡萄糖转运蛋白4(GLUT4)和钠-葡萄糖协同转运蛋白2(SGLT2)为关键靶点,系统性地筛选与发现具备潜在抗糖尿病活性的化合物。具体而言,研究内容主要围绕天然产物资源库,深入探索针对GLUT4与SGLT2靶点的抗糖尿病活性成分及其内在作用机制。同时,研究也借助高分辨傅里叶变换质谱技术,定向发现能够抑制相关酶活性的小分子抑制剂。在基于天然产物的先导化合物发现流程中,首先系统性地建立了涵盖传统中草药及民族药物的实物样品库。随后,依据药物研发中经典的Lipinski五规则,通过采用特殊的柱色谱前处理技术,构建了一个主要富集先导化合物类似组分的微量提取物库。为进一步扩大筛选范围,研究还开发了一种独特的柱色谱技术,用以高效捕捉与先导化合物或已知药物分子结构相似的天然产物。应用此项技术,在已有的上千种微量提取物基础上,成功构建了一个包含超过五千个预分离组分的化合物库,其中每个组分均可能含有先导化合物类似物或具有新药开发潜力的天然产物分子。在针对GLUT4靶点的抗糖尿病化合物发现方面,研究首先明确了GLUT4作为生物体内调节葡萄糖摄取与利用的核心蛋白的重要地位。在此基础上,建立了一套基于GLUT4靶点的体外抗糖尿病活性筛选体系。进而,利用Ⅱ型糖尿病动物模型,对筛选出的活性化合物进行了系统的体内药效学评价。最后,通过免疫印迹等分子生物学技术,深入揭示了这些活性化合物发挥抗糖尿病作用的具体分子机制与信号通路。

在学术成果方面,本人曾参与多项专业著作的编纂工作。具体包括:其一,担任《中国五峰特色常见药用植物》的编委,该书于2014年9月正式出版,为第一版;其二,在《湖北中药志》的编纂过程中,亦担任编委职务;其三,作为第一副主编,深度参与了《中国毒性民族药志》的编写与审定,该书于2015年11月由科学出版社发行,同样为第一版。这些经历系统锻炼了本人在专业领域的组织、撰写与编辑能力。
